After a meet and great, when do you hit the book it now button ?

I always let the client book the stay, especially if it is a new client. If they are new to rover, I explain how the booking process works and then say that I will let them make the decision. I emphasize (when appropriate) that I would love to keep their dog and to let me know if they have any further questions.

If they don't book within 24 hours, I send a reminder message asking if they still want me to keep their dog. That way, they know I'm still interested and available.

By the end of the M&G, both you and the client should have a pretty good idea whether or not you want to book the stay. I'll tell the owner that I'd love to watch their dog, and if they agree, I'll explain the rest of the process and tell them I'll press the book it now button as soon as they leave, so usually the stay is confirmed shortly after they get home. Occasionally, if the owner has other M&Gs scheduled, I'll tell them I'd love to watch their dog, and just ask that they let me know what they've decided after their other meetings are complete.

That depends on how the meeting went. Did they mention they were meeting other potential sitters or did they say they definitely want to book with you? My experience has been after the client has gone home, at some point, they start the booking process. I edit rates if necessary, add messages confirming the details, then accept. If it's a first time Rover client, they have to input their payment info. first. So, for my first booking with a new client, they initiate the process. Maybe other sitters have different experiences.
